大鼠模型是生物醫(yī)學(xué)研究中重要的資源,因與人類免疫系統(tǒng)相似,而常被用于免疫相關(guān)的功能研究。然而目前小鼠模型在基礎(chǔ)和機(jī)制免疫學(xué)研究中占主導(dǎo)地位,相對(duì)缺乏對(duì)大鼠模型中免疫細(xì)胞的流式分析鑒定。本文將分享對(duì)大鼠常見免疫細(xì)胞的流式鑒定方案,鑒定分析不同組織中的粒細(xì)胞、巨噬細(xì)胞、T細(xì)胞、B細(xì)胞及NK細(xì)胞。
實(shí)驗(yàn)方案1
該方案以10-12周齡Lewis大鼠的肌肉、脾臟、胸腺和腸系膜淋巴結(jié)為樣本,組織經(jīng)消化并用40μm細(xì)胞過濾器過濾后制備成單細(xì)胞懸液。

圈門分析:
1. 側(cè)向散射(SSC)和前向散射(FSC)參數(shù)對(duì)樣本細(xì)胞進(jìn)行圈門,利用FSC-H去除粘連細(xì)胞,然后進(jìn)行單一細(xì)胞圈門;采用死活染料及CD45對(duì)活性良好的CD45+細(xì)胞進(jìn)行下游分析;
2. 巨噬細(xì)胞:根據(jù)CD11B、CD68圈出巨噬細(xì)胞(CD11B+CD68+),再根據(jù)CD86和CD163分別圈出M1(CD86+)和M2(CD163+)型巨噬細(xì)胞;
3. 單核細(xì)胞:根據(jù)CD11B、CD43圈出單核細(xì)胞(CD11B+CD43+);
4. 粒細(xì)胞:根據(jù)CD11B、CD43、CD62L圈出粒細(xì)胞(CD11B+CD43+CD62L+/-SSChi),當(dāng)粒細(xì)胞被激活時(shí)CD62L則低表達(dá);
5. 淋巴細(xì)胞:利用TCR?δ圈出?δT細(xì)胞,接著在TCR?δ-的細(xì)胞中,進(jìn)一步根據(jù)CD3劃分 T 細(xì)胞(CD3+)和非T(CD3-)淋巴細(xì)胞;
(1)T細(xì)胞:在CD3+T細(xì)胞中,根據(jù)CD4、CD8圈出CD4+和CD8+ T細(xì)胞;在CD4+T細(xì)胞中圈出Treg細(xì)胞(CD25+)、Th1細(xì)胞(CCR1+CCR5+)和的Th2細(xì)胞(CCR4+CCR8+);
在CD3+T細(xì)胞中,在根據(jù)CD44和CD62L圈出激活性T細(xì)胞(CD44+CD62L-)、記憶T細(xì)胞(CD44hiCD62Llo)、naive T細(xì)胞(CD44loCD62Lhi);
(2)B細(xì)胞:非T細(xì)胞淋巴細(xì)胞中,根據(jù)CD45R圈出B細(xì)胞(CD3-CD45R+);
(3)NK細(xì)胞:非T細(xì)胞淋巴細(xì)胞中,根據(jù)CD49b圈出NK細(xì)胞(CD3-CD49b-)。
實(shí)驗(yàn)方案2
該方案以Sprague-Dawley大鼠的血液和肺、脾臟、肝臟、骨髓細(xì)胞為樣本。

1. 側(cè)向散射(SSC)和前向散射(FSC)參數(shù)對(duì)樣本細(xì)胞進(jìn)行圈門,去除碎片(i),利用死活染料去除死細(xì)胞(ii),利用FSC-H和FSC-W參數(shù)來排除粘連細(xì)胞(iii),利用CD45圈出淋巴細(xì)胞(iv);
2. T細(xì)胞:從CD45+細(xì)胞中,圈出CD3+T細(xì)胞(v),根據(jù)CD4、CD8圈出CD4+T和CD8+ T細(xì)胞(vi);
3. NK細(xì)胞;從CD45+CD3-的細(xì)胞中,圈出CD161+His48-細(xì)胞(vii);
4. B細(xì)胞:從CD45+CD3-CD161-His48+/-的細(xì)胞,圈出CD45R+CD161-細(xì)胞(viii);
5. 中性粒細(xì)胞:從CD45+CD3-CD161-CD45R-的細(xì)胞中,圈出CD43+細(xì)胞(ix);
6. 單核細(xì)胞:從CD45+CD3-CD161-CD45R-CD43-/lo的細(xì)胞中,利用CD43和His48的差異表達(dá)圈出兩個(gè)單核細(xì)胞亞群 ——CD43LoHis48Hi 和 CD43HiHis48Lo-Int(x)。
總結(jié)
兩個(gè)方案為大鼠常見免疫細(xì)胞的鑒定分析提供了框架,但對(duì)一些細(xì)胞群的鑒定有所差異:方案1通過CD45+CD3-CD49B+鑒定NK細(xì)胞,方案2則通過CD45+CD3-CD163-鑒定NK細(xì)胞;方案1通過CD45+CD11B+CD43+鑒定單核細(xì)胞,而方案2通過CD43和His48將單核細(xì)胞分為兩群(CD45+CD3-CD161-CD45R-CD43-/loCD43LoHis48Hi 、 CD45+CD3-CD161-CD45R-CD43-/loCD43HiHis48Lo-Int),強(qiáng)調(diào)了CD43和His48指標(biāo)對(duì)中性粒和單核細(xì)胞鑒定中的實(shí)用性。
